Sustrato sensible de peroxidasa fluorogénica Amplite® Red es una de las sondas estables para detectar HRP y H2O2.

Descripción

Nuestro sustrato sensible de peroxidasa fluorogénica Amplite® Red genera un producto fluorescente muy rojo que tiene una absorción máxima de 571 nm y una emisión máxima de 585 nm.

A diferencia de otros sustratos de HRP, como las dihidrofluoresceínas y las dihidrorodaminas, la oxidación por aire de Amplite® Red es mínima. Amplite® Red es una de las sondas fluorogénicas más sensibles y estables para detectar HRP y H2O2. Amplite® Red se ha utilizado ampliamente para detectar HRP en muchos inmunoensayos. Por otro lado, Amplite® Red también se puede utilizar para detectar trazas de H2O2. La detección de H2O2 basada en Amplite® Red es al menos un orden de magnitud más sensible que el ensayo de escopoletina comúnmente usado para H2O2.

Debido a que el H2O2 se produce en muchas reacciones enzimáticas redox, Amplite® Red puede usarse en reacciones enzimáticas acopladas para detectar la actividad de muchas oxidasas y/o enzimas/sustratos o cofactores relacionados, como glucosa, acetilcolina y colesterol, L-glutamato, aminoácidos , etc.

Preparación de Soluciones de Stock

A menos que se indique lo contrario, todas las soluciones madre no utilizadas deben dividirse en alícuotas de un solo uso y almacenarse a -20 °C después de la preparación. Evite los ciclos repetidos de congelación y descongelación.

Solución madre de AmpliteTM Red (250X):
Agregue 200 ml de DMSO anhidro en el vial y mezcle bien. La solución madre debe usarse con prontitud. Cualquier solución no utilizada debe dividirse en alícuotas y volver a congelarse a < -20 oC. Nota: Evite los ciclos repetidos de congelación y descongelación y protéjalo de la luz.


Preparación de Soluciones de Trabajo

  1. Solución de trabajo de peroxidasa roja AmpliteTM (1X):
    Agregue 20 μL de solución madre AmpliteTM Red (250X) en 5 mL de buffer de fosfato 50 mM o el buffer de su elección, pH 7 con H2O2 200 mM. Nota: AmpliteTM Red es inestable en presencia de tioles como DTT y b-mercaptoetanol. Los tioles superiores a 10 μM (concentración final) podrían disminuir significativamente el rango dinámico del ensayo. El NADH y el glutatión (reducido de: GSH) pueden interferir con el ensayo
  2. Solución de trabajo AmpliteTM Red H2O2 (1X):
    Agregue 20 μL de solución madre AmpliteTM Red (250X) en 5 mL de buffer de fosfato 50 mM o el buffer de su elección, pH 7 con 0,4 unidades/mL de peroxidasa. Nota: AmpliteTM Red es inestable en presencia de tioles como DTT y b-mercaptoetanol. Los tioles superiores a 10 μM (concentración final) podrían disminuir significativamente el rango dinámico del ensayo. El NADH y el glutatión (reducido de: GSH) pueden interferir con el ensayo.
